Magruder was probably born in Allegany County, Maryland. The place of his marriage to Margaret Porter is not known, but it could have taken place either in Brooke County, (West) Virginia, or in Allegany County, Maryland. John W. and Margaret Porter Magruder are buried in the Milledgeville-Plymouth Cemetery near Milledgeville, Fayette County, Ohio. The John W. Magruder family was living in Allegany County, Maryland, in 1830. By 1840, they had moved to (West) Virginia and settled in Brooke County. They moved to Ohio County, (West) Virginia, by 1850, and lived there until about 1870, when they moved to Clinton County, Ohio. After two or three years in Clinton County, they moved to Fayette County, and settled near Milledgeville.
John W. Magruder appears in a list of names in the (Wheeling, West Virginia) Daily Intelligencer of April 25, 1860. The list appears to be a list of voters who are being asked to attend a meeting in Wheeling on 2 May 2 1860 to elect delegates to the Republican national convention.
On 29 April 1879 John W. Magruder sold 12 acres of land in Fayette County, Ohio, to his son, Joshua L. Magruder, for $800.00 (Book 4, page 504, Survey 974). On 8 August 1879 Joshua L. Magruder sold 15 acres to John W. Magruder in Fayette County, Ohio (Book 4, page 313, Survey 974). On the same day, John sold the land to Thomas Dawson for $850 (Book 4, page 313, Survey 974). Thomas Dawson may have been a relative of John's mother, Ann Dawson Magruder. Thomas sold the land to Marcus Barclay on 2 March 1880.

The Bible was first known to have belonged to Hester Caroline Magruder Howell, although she could have received it from hjer father, John W. Magruder. Hester Magruder Howell gave the Bible to her daughter, Ida Howell Hyer, who in turn gave it to John Garrett Magruder.
